Anti-GPC3-CAR autologous T lymphocytes are a form of **chimeric antigen receptor (CAR) T cell therapy** in which a patient’s own T cells are genetically engineered to express a CAR that specifically targets **glypican-3 (GPC3)**, a protein highly expressed in hepatocellular carcinoma (HCC) and rarely in normal tissues. The therapy involves isolating T cells from the patient, modifying them ex vivo to express the anti-GPC3 CAR (often using lentiviral vectors), expanding them, and then reinfusing them into the patient after a lymphodepleting chemotherapy regimen. The modified T cells recognize and destroy GPC3-expressing tumor cells, leveraging the immune system for targeted anti-tumor activity. This approach is being developed primarily for **advanced HCC** not amenable to curative treatment and has shown promising safety and early anti-tumor activity in early-phase clinical trials. Trials aim to determine the optimal dose, safety, feasibility, efficacy, and persistence of these engineered T cells in patients[1][2][4][6][7].
