Overview

anti-GPC3 CAR-T cells (Xinqiao Hospital) is an autologous chimeric antigen receptor (CAR) T-cell therapy designed to target Glypican-3 (GPC3), an oncofetal antigen highly expressed in hepatocellular carcinoma (HCC) but with limited expression in normal adult tissues. The CAR construct utilized by Xinqiao Hospital consists of an anti-GPC3 single-chain variable fragment (scFv) for antigen recognition, a 4-1BB (CD137) costimulatory domain to enhance T-cell persistence and metabolic fitness, and a CD3ζ signaling domain for T-cell activation. Additionally, the construct incorporates a truncated epidermal growth factor receptor (tEGFR), which serves as a selection marker during manufacturing and a potential safety switch for antibody-mediated depletion. In clinical trials, this therapy is often administered following lymphodepleting chemotherapy (fludarabine and cyclophosphamide) and in combination with transcatheter arterial chemoembolization (TACE) to treat patients with advanced, GPC3-positive HCC.
