Drug intelligence / Profile preview
An anti-GPNMB monoclonal antibody is a therapeutic antibody designed to target glycoprotein nonmetastatic melanoma protein B (GPNMB), a type I transmembrane glycoprotein. GPNMB is highly expressed in various cancers (such as melanoma and triple-negative breast cancer) and is also upregulated on specific macrophage subpopulations (such as C1Q+/GPNMB+/CD68+ macrophages) in fibrotic tissues [1]. In the context of chronic liver disease, particularly hepatitis B virus (HBV)-associated liver fibrosis, GPNMB-positive macrophages drive fibrotic progression [1]. Targeting GPNMB with a monoclonal antibody has been shown preclinically to suppress hepatic fibrosis and reduce the expression of fibrosis-associated genes [1]. Additionally, anti-GPNMB antibodies have been investigated in oncology, both as bare antibodies and as the targeting component of antibody-drug conjugates (such as glembatumumab vedotin), and are being explored for other indications including obesity, atherosclerosis, and schistosomiasis-induced fibrosis.
