Drug intelligence / Profile preview
Anti-GPR133 antibody refers to antibodies targeting GPR133 (ADGRD1), an adhesion G protein-coupled receptor (aGPCR) that is de novo expressed in glioblastoma (GBM) and is necessary for tumor growth. GPR133 undergoes autoproteolytic cleavage into an extracellular N-terminal fragment (NTF) and a membrane-bound C-terminal fragment (CTF). Antibodies targeting the N-terminus (such as the mouse monoclonal clone 8E3E8) bind to the pentraxin (PTX) domain of the NTF, promoting NTF-CTF dissociation and enhancing canonical Gs-mediated signaling, which increases cytosolic cAMP. This modulation of GPR133 function serves as a proof-of-concept for developing biologics to target glioblastoma.
