Overview

The anti-GPR133 NTF antibody is an experimental research-stage biologic designed to target the N-terminal fragment (NTF) of GPR133 (also known as ADGRD1), an adhesion G protein-coupled receptor. GPR133 is de novo expressed in glioblastoma (GBM) relative to healthy brain tissue and is essential for tumor growth, making it a promising therapeutic target. These antibodies modulate GPR133 signaling—specifically increasing activation via Gs coupling and subsequent cytosolic cAMP elevation—by promoting the dissociation of the cleaved NTF from the C-terminal fragment (CTF) or by altering the flexibility of the GAIN domain. This proof-of-concept paradigm demonstrates the potential for using biologics to modulate adhesion GPCR function in the context of neuro-oncology.
