Anti-GPR77 antibody is a monoclonal antibody targeting G-protein coupled receptor 77 (GPR77), also known as complement C5a receptor 2 (C5AR2) or C5L2. GPR77 is a receptor for the complement anaphylatoxin C5a. In the tumor microenvironment, a specific subset of cancer-associated fibroblasts (CAFs) characterized by CD10 and GPR77 expression promotes cancer stemness, tumor formation, and chemoresistance. Mechanistically, complement signaling via GPR77 maintains persistent NF-kappaB activation in these CAFs, which in turn sustains a survival niche for cancer stem cells. Neutralizing anti-GPR77 antibodies block this signaling pathway, reducing the infiltration of GPR77+ CAFs, abolishing tumor formation, and restoring chemosensitivity to chemotherapeutic agents such as docetaxel in preclinical models of breast and lung cancer.
