Anti-GPRC5D bispecific T-cell engagers (TCEs) are a class of immunotherapy designed to treat multiple myeloma by simultaneously binding to G protein-coupled receptor class C group 5 member D (GPRC5D) on malignant plasma cells and the CD3 receptor on T cells. This dual binding facilitates the recruitment and activation of T cells, leading to the direct lysis of the GPRC5D-expressing tumor cells. GPRC5D is highly and specifically expressed on plasma cells, making it an ideal target for multiple myeloma therapies, particularly in patients who have relapsed or are refractory to other treatments like BCMA-targeted therapies. Notable examples in this class include talquetamab and forimtamig.
