Overview

The anti-GPRC5D CAR-T therapy developed by the 920th Hospital of Joint Logistics Support Force is a second-generation chimeric antigen receptor (CAR) T-cell product designed to treat relapsed or refractory multiple myeloma. The construct features a humanized rabbit monoclonal antibody-derived single-chain variable fragment (scFv) targeting G protein-coupled receptor class C group 5 member D (GPRC5D), a protein highly expressed on malignant plasma cells. This scFv is linked to a 4-1BB costimulatory domain and a CD3ζ signaling domain to enhance T-cell activation and persistence. Clinical evaluation in investigator-initiated trials (NCT05739188) has demonstrated promising efficacy with high overall response rates and a manageable safety profile in patients who have failed prior lines of therapy.
