Drug intelligence / Profile preview
An anti-Gremlin1 neutralizing antibody is a therapeutic monoclonal antibody designed to target and neutralize Gremlin-1 (GREM1), a highly conserved secreted glycoprotein and member of the DAN family of bone morphogenetic protein (BMP) antagonists. Gremlin-1 is highly expressed by stromal cells, cancer-associated fibroblasts (CAFs), and tumor cells in various human carcinomas (such as colorectal, prostate, pancreatic, and breast cancers), where it represses BMP signaling (specifically BMP-2, BMP-4, and BMP-7) to promote epithelial-mesenchymal transition (EMT), lineage plasticity, and treatment resistance. By binding to Gremlin-1, these neutralizing antibodies block its interaction with BMPs, thereby restoring BMP signaling pathways and downstream SMAD activation, which inhibits tumor cell growth, proliferation, and metastasis. Key clinical-stage candidates in this class include Transcenta's TST003 and UCB's ginisortamab (UCB6114), which are being evaluated for the treatment of advanced solid tumors, including castration-resistant prostate cancer and colorectal cancer. Additionally, preclinical research suggests potential applications in multiple myeloma, ocular angiogenic diseases, and pulmonary arterial hypertension.
