Overview

Anti-GUCY2C CAR-T cells are a nanobody-based chimeric antigen receptor (CAR) T-cell therapy designed to target Guanylate Cyclase 2C (GUCY2C), a transmembrane receptor primarily expressed on the apical membrane of intestinal epithelial cells and frequently overexpressed in colorectal cancer (CRC). Developed by Parasol Biotechnology in collaboration with the University of Science and Technology of China, this autologous cell therapy utilizes a nanobody-derived antigen-binding domain to enhance specificity and potentially reduce immunogenicity compared to traditional scFv-based CARs. In early-phase clinical trials (such as ChiCTR2100044831), these CAR-T cells have demonstrated a manageable safety profile and promising objective response rates in patients with heavily pretreated metastatic colorectal cancer.
