The anti-HBsAg nanobody 2nd generation is a therapeutic single-domain antibody (VHH) designed to target the Hepatitis B surface antigen (HBsAg). Developed by the National Institute of Diagnostics and Vaccine Development in Infectious Diseases (NIDVD) at Xiamen University, this agent is intended for the treatment of chronic hepatitis B. It functions by binding to HBsAg to neutralize the virus and facilitate the clearance of circulating surface antigens, which is a critical step toward achieving a functional cure. The "second-generation" designation typically refers to an optimized structure, such as a biparatopic or multivalent format (e.g., VHH-003), providing enhanced binding affinity and broader coverage of HBV genotypes compared to earlier monovalent versions. It is currently being evaluated in Phase 1 clinical trials to assess its safety, tolerability, and preliminary efficacy in reducing HBsAg levels.
