Overview

The anti-hCD33 monoclonal antibody (specifically clone 4L3-46) is an antagonistic biologic developed by researchers at Academia Sinica that targets human CD33 (also known as Siglec-3). CD33 is an inhibitory receptor containing an immune-receptor tyrosine-based inhibition motif (ITIM) that typically recruits phosphatases such as SHP-1, SHP-2, and SHIP to exert immunosuppressive effects. While CD33 is a well-established therapeutic target in acute myeloid leukemia (AML) and serves as a marker for myeloid-derived suppressor cells (MDSCs), this specific antibody is being investigated for its potential to treat solid tumors. By blocking CD33 signaling, 4L3-46 aims to restructure the infiltrating MDSC population and reverse CD33-mediated immune evasion, thereby attenuating tumor growth and improving survival outcomes in preclinical models.
