Overview

An anti-hemojuvelin monoclonal antibody is a biologic therapeutic designed to target hemojuvelin (HJV), also known as repulsive guidance molecule c (RGMc). HJV is a bone morphogenetic protein (BMP) co-receptor that plays a pivotal role in the regulation of hepcidin, the master hormone of systemic iron homeostasis. By binding to HJV, the antibody inhibits the BMP/SMAD signaling pathway that induces hepcidin expression in the liver. In conditions of chronic inflammation, such as inflammatory bowel disease (IBD), pathologically high hepcidin levels lead to iron sequestration and anemia of chronic disease. Neutralizing HJV reduces hepcidin levels, thereby mobilizing iron stores, increasing serum iron availability, and stimulating erythropoiesis. Beyond treating anemia, these antibodies are being investigated for potential disease-modifying effects in inflammatory disorders by modulating iron-dependent pathways and inflammatory signaling. The lead candidate in this class, FE-201 (formerly h5F9-AM8), is being developed by Ferrumax Pharmaceuticals.
