"anti-HER2 ADC 7" is an experimental, homogeneous antibody-drug conjugate (ADC) developed by AbTis Co., Ltd. for the treatment of HER2-expressing tumors, specifically glioblastoma multiforme (GBM). It consists of the anti-HER2 monoclonal antibody trastuzumab site-specifically conjugated to the microtubule inhibitor monomethyl auristatin E (MMAE) via a non-cleavable maleimidocaproyl (mc) linker, achieving a drug-to-antibody ratio (DAR) of 4.0. The conjugate is produced using AbTis's proprietary AbClick technology, which enables site-specific conjugation to the Fc region of native antibodies without genetic engineering. Research published in 2022 highlighted ADC 7 in studies comparing the blood-brain barrier (BBB) penetration and therapeutic efficacy of homogeneous versus heterogeneous ADCs, demonstrating that homogeneous conjugates with optimal payload loading provide superior brain tumor targeting and survival benefits in orthotopic models.
