Anti-HER2 CAR T-cells (NCI) is an autologous cell therapy developed by the National Cancer Institute's Surgery Branch for the treatment of metastatic cancers overexpressing the HER2 protein. The therapy involves the genetic modification of a patient's peripheral blood lymphocytes (PBLs) using a retroviral vector to express a chimeric antigen receptor (CAR) specific for HER2. The treatment protocol typically includes a lymphodepleting conditioning regimen of cyclophosphamide and fludarabine to eliminate regulatory T cells and competing lymphocytes, followed by the infusion of the engineered CAR T-cells. High-dose aldesleukin (interleukin-2) is subsequently administered to support the expansion and persistence of the infused cells in vivo. This approach aims to direct the patient's immune system to recognize and destroy HER2-positive tumor cells across various solid tumor types.
