Anti-HER2 IgE is an experimental monoclonal antibody of the immunoglobulin E (IgE) class engineered to target the human epidermal growth factor receptor 2 (HER2/neu), an oncoprotein frequently overexpressed in breast and ovarian cancers. Developed by Professor Manuel Penichet at the University of California, Los Angeles (UCLA) and licensed to Quest PharmaTech (through its subsidiary OncoQuest), this therapeutic combines the variable regions of a HER2-specific antibody with the constant region of a human IgE antibody. Unlike conventional IgG-based therapies, this IgE-based approach leverages the exceptionally high affinity of IgE for FcεRI receptors on effector cells such as mast cells, basophils, and macrophages. This engagement triggers potent antibody-dependent cellular cytotoxicity (ADCC), degranulation, and enhanced antigen presentation, potentially overcoming resistance to standard IgG-based anti-HER2 treatments and reprogramming the tumor microenvironment toward a pro-inflammatory, anti-tumor state.
