An anti-HER3 ADC (Antibody-Drug Conjugate) is a targeted cancer therapeutic designed to deliver a cytotoxic payload directly to cells expressing the Human Epidermal Growth Factor Receptor 3 (HER3/ERBB3). HER3 is a member of the ErbB/HER receptor tyrosine kinase family and is frequently overexpressed or activated in various solid tumors, including non-small cell lung cancer (NSCLC), breast cancer, and colorectal cancer, where it contributes to tumor growth, survival, and resistance to other targeted therapies. The ADC typically comprises a humanized monoclonal antibody targeting the extracellular domain of HER3, a stable or cleavable chemical linker, and a potent cytotoxic agent such as a topoisomerase I inhibitor or a microtubule inhibitor. Upon binding to HER3, the complex is internalized via endocytosis, and the payload is released within the lysosomal compartment, leading to DNA damage or mitotic arrest and subsequent cell death. Several anti-HER3 ADCs are currently in clinical development, with patritumab deruxtecan (U3-1402) being the most clinically advanced candidate.
