Drug intelligence / Profile preview
The anti-HK antibody (specifically clone 3E8) is a monoclonal antibody that targets domain 6 (D6) of high-molecular-weight kininogen (HK, encoded by the KNG1 gene). Developed by researchers at The Rockefeller University, the antibody binds with high affinity to HK, preventing its cleavage and the subsequent release of the inflammatory mediator bradykinin. Mechanistically, 3E8 blocks the assembly of the trimolecular contact activation complex by inhibiting the binding of prekallikrein (PK) and factor XI (FXI) to HK, thereby suppressing both the inflammatory and intrinsic coagulation pathways of the plasma contact system. It is being investigated preclinically for the treatment of neuroinflammatory and vascular pathologies, including Alzheimer's disease and multiple sclerosis (MS) / experimental autoimmune encephalomyelitis (EAE).
