Drug intelligence / Profile preview
Anti-HLA-A2 CAR-Tregs are regulatory T cells (Tregs) genetically engineered to express a chimeric antigen receptor (CAR) targeting human leukocyte antigen A2 (HLA-A2). This cell therapy platform is designed to induce localized immunological tolerance and prevent transplant rejection in HLA-A2 mismatched solid organ transplantations (where an HLA-A2 negative recipient receives an HLA-A2 positive donor organ). Upon administration, the CAR-Tregs home to the transplanted organ, where they are activated by the donor's HLA-A2 antigen to suppress local inflammatory and alloimmune responses. This targeted approach aims to protect the graft while reducing or eliminating the need for chronic, systemic, and nephrotoxic immunosuppressive regimens. Key clinical-stage candidates under this platform include Quell Therapeutics' QEL-001, currently in Phase 1/2 development for liver transplantation, and Sangamo Therapeutics' TX200, in Phase 1/2 development for kidney transplantation.
