Anti-HMGB1 monoclonal antibody is a laboratory-engineered monoclonal antibody specific for high-mobility group box 1 (HMGB1), a chromatin-associated protein released extracellularly during cellular stress and damage[1][3][5][6]. By binding to HMGB1, these antibodies neutralize its activity, thereby reducing pro-inflammatory signaling and tissue damage in various animal models of central nervous system and peripheral inflammatory diseases—including stroke, spinal cord ischemia-reperfusion injury, traumatic brain injury, epilepsy, Alzheimer's disease, Parkinson's disease, and trauma-induced immunosuppression[2][3][4][5][6]. Mechanism of action involves direct binding and neutralization of extracellular HMGB1, blocking its interaction with receptors such as toll-like receptor 4 (TLR4) and receptor for advanced glycation endproducts (RAGE), leading to decreased neuroinflammation, blood-brain barrier protection, and reduced immune cell infiltration[2][4][6]. Developed and manufactured primarily for research purposes (not clinical human use), examples include the DPH1.1 clone generated by immunizing mice with HMGB1 peptide[1].
