anti-hOX40 Ab2 is a novel agonistic monoclonal antibody that targets human OX40 (also known as CD134 or TNFRSF4), a co-stimulatory member of the tumor necrosis factor receptor (TNFR) superfamily. OX40 is primarily expressed on the surface of activated T cells, including effector T cells and regulatory T cells (Tregs). The binding of Ab2 to OX40 mimics the action of its natural ligand (TNFSF4), leading to the expansion of effector T cells, increased cytokine release, and enhanced survival of antigen-specific memory T cells. In preclinical research presented at AACR 2024 by the Shanghai Model Organisms Center, Ab2 demonstrated potent anti-tumor activity as a monotherapy in humanized OX40 mouse models of pancreatic and colorectal cancer. The antibody was also studied in various combination regimens with PD-1 inhibitors to investigate the impact of dosing sequence and timing on therapeutic efficacy.
