anti-hOX40 antibody 1 is a human-specific agonistic monoclonal antibody targeting OX40 (CD134), a member of the tumor necrosis factor receptor superfamily (TNFRSF4). OX40 is primarily expressed on activated T cells, and its activation provides a potent costimulatory signal that enhances effector T cell proliferation, survival, and cytokine production while potentially suppressing regulatory T cell (Treg) activity. In preclinical research presented at AACR 2024, this antibody (referred to as Ab1) demonstrated dose-dependent anti-tumor efficacy in humanized OX40 knock-in mouse models, particularly in syngeneic MC38 tumor models. The study evaluated its performance as a monotherapy and in combination with PD-1 inhibitors to determine optimal dosing sequences for immunotherapy regimens.
