Overview

Anti-hSIGIRR antibody is a monoclonal antibody designed to target and block the Single Ig IL-1-related receptor (SIGIRR), also known as IL-1R8 or TIR8. IL-1R8 is a member of the interleukin-1 receptor family that serves as a potent negative regulator of the IL-1R and Toll-like receptor (TLR) signaling pathways. In the tumor microenvironment, IL-1R8 acts as an immune checkpoint, particularly on natural killer (NK) cells and T cells, suppressing their activation and antitumor activity. By antagonizing IL-1R8, this antibody releases these inhibitory signals, thereby enhancing the recruitment, maturation, and effector functions (such as IFN-gamma production and cytotoxicity) of NK cells against malignant cells. This approach is being explored as a novel immunotherapy for solid tumors, including colorectal cancer liver metastases, where traditional checkpoint inhibitors often show limited efficacy.
