Anti-IARS neutralizing antibody is an experimental monoclonal antibody that targets isoleucyl-tRNA synthetase (IARS), an enzyme traditionally involved in protein synthesis that has been found to have non-canonical pro-inflammatory roles in the skin. In the context of psoriasis, IARS is upregulated in epidermal keratinocytes and promotes inflammation via the Jak2/STAT3/CXCL16 signaling pathway. By binding to and neutralizing IARS, the antibody inhibits this pathway, leading to decreased expression of chemokines and cytokines such as IL-6, IL-1β, and S100A7. Preclinical research in imiquimod-induced psoriasiform mouse models has demonstrated that administration of these neutralizing antibodies can significantly reduce epidermal thickness and inflammatory cell infiltration, suggesting a potential new therapeutic strategy for treating psoriasis.
