Overview

Anti-IDO2 siRNA is a small interfering RNA (siRNA) therapeutic designed to silence the expression of indoleamine 2,3-dioxygenase 2 (IDO2). Unlike its counterpart IDO1, which is primarily immunosuppressive, IDO2 promotes inflammation through a B-cell intrinsic mechanism. This specific therapeutic approach utilizes a targeted DNA nanostructure-based delivery system to specifically inhibit IDO2 function in B cells. Preclinical studies in murine models of arthritis, such as the KRN.g7 model and collagen-induced arthritis, have demonstrated that silencing IDO2 reduces autoantibody production and attenuates joint inflammation, mimicking the phenotype of genetic knockouts. Research also indicates that IDO2 expression is elevated in B cells from patients with rheumatoid arthritis (RA), and in vitro studies have shown that this expression can be effectively silenced using anti-IDO2 siRNA, suggesting potential for human therapeutic application in autoimmune diseases.
