Anti-IFNα antibodies are a class of monoclonal antibodies designed to neutralize interferon-alpha (IFNα), a type I interferon that plays a central role in the pathogenesis of various autoimmune diseases, most notably systemic lupus erythematosus (SLE). By binding directly to multiple subtypes of IFNα, these antibodies prevent the cytokine from interacting with its receptor (IFNAR), thereby inhibiting the downstream signaling pathways that lead to the activation of the innate and adaptive immune systems. In preclinical models, such as the Trex1 knockout mouse, anti-IFNα treatment has demonstrated efficacy in improving survival and reducing organ damage, such as leukocyte infiltration in the heart. Clinical candidates in this space have included sifalimumab and rontalizumab, though recent focus has shifted toward targeting the receptor (IFNAR) itself (e.g., anifrolumab).
