Overview

Anti-IFNγ refers to a class of therapeutic agents, primarily monoclonal antibodies, designed to neutralize interferon-gamma (IFNγ), a potent pro-inflammatory cytokine. IFNγ is a key mediator of innate and adaptive immunity, but its overproduction is implicated in the pathogenesis of various autoinflammatory and autoimmune diseases. By binding directly to IFNγ, these agents prevent its interaction with the IFNγ receptor complex (IFNGR1/IFNGR2), thereby inhibiting downstream signaling through the JAK-STAT pathway. The most prominent clinical representative of this class is emapalumab (Gamifant), which is FDA-approved for the treatment of primary hemophagocytic lymphohistiocytosis (HLH). In research settings, anti-IFNγ antibodies are frequently employed to study cytokine-driven tissue injury, such as in models of intestinal epithelial cell death and acute enteropathy, where they have been shown to abrogate inflammation and promote cell survival.
