Anti-IFNAR1 antibodies are a class of therapeutic agents, primarily monoclonal antibodies, that target the subunit 1 of the Type I interferon receptor (IFNAR1). By binding to IFNAR1, these antibodies prevent the formation of the active ternary signaling complex with IFNAR2 and Type I interferons (such as IFN-α, IFN-β, and IFN-ω), thereby inhibiting the downstream JAK-STAT signaling pathway. This blockade reduces the expression of interferon-stimulated genes (ISGs), which are often elevated in various autoimmune and inflammatory conditions. While anifrolumab is the most prominent clinical-stage example approved for systemic lupus erythematosus, research continues into the use of IFNAR1 inhibition for other conditions characterized by high interferon signatures, including lichenoid chronic graft-versus-host disease (lGVHD) and other dermatomyositis-related skin inflammations.
