An anti-IFNGR1 blocking antibody is a therapeutic monoclonal antibody designed to target and inhibit the interferon-gamma receptor 1 (IFNGR1), also known as the alpha chain of the Type II interferon receptor (CD119). By binding to IFNGR1, the antibody competitively prevents the pro-inflammatory cytokine interferon-gamma (IFN-γ) from associating with its receptor complex, thereby blocking the activation of the downstream JAK/STAT signaling pathway, specifically involving JAK1, JAK2, and STAT1. IFN-γ is a potent mediator of Th1-type immune responses and plays a central role in the pathogenesis of various autoimmune and hyperinflammatory conditions. While clinical development in this pathway has largely focused on neutralizing the IFN-γ ligand itself (e.g., emapalumab), direct receptor blockade offers an alternative pharmacological strategy to dampen excessive interferon signaling. Potential therapeutic applications for such agents include the treatment of hemophagocytic lymphohistiocytosis (HLH), systemic lupus erythematosus (SLE), and other interferon-mediated inflammatory disorders.
