Overview

"anti-IGHV1-69 CAR T cells" is a novel Chimeric Antigen Receptor (CAR) T cell immunotherapy designed to specifically target B cells expressing the immunoglobulin heavy-chain variable region IGHV1-69. This targeted approach aims to treat B cell-mediated autoimmune diseases, such as immune-mediated thrombotic thrombocytopenic purpura (iTPP), by selectively depleting pathogenic IGHV1-69+ B cells responsible for autoantibody production (e.g., anti-ADAMTS13 autoantibodies in iTPP) while largely sparing the normal B cell repertoire. The CAR construct, named CAR1-69, utilizes a CD8-41BB-CD3z signaling domain and a single-chain variable fragment (scFv) derived from the humanized murine G6.3 monoclonal antibody to recognize the IGHV1-69+ B-cell receptor. This specificity is intended to mitigate the B cell aplasia and hypogammaglobulinemia often associated with broader CD19-directed CAR T cell therapies.
