anti-IGSF8 antibody refers to a class of monoclonal antibodies designed to target Immunoglobulin Superfamily Member 8 (IGSF8), a novel innate immune checkpoint. The most prominent clinical-stage representative is GV20-0251, developed by Gv20 Therapeutics and Shanghai Xunbaihui Biotechnology. IGSF8 is frequently overexpressed on the surface of various cancer cells and interacts with the inhibitory receptor KIR3DL2 on natural killer (NK) cells, thereby suppressing the immune system's ability to eliminate tumors. By blocking the IGSF8-KIR3DL2 interaction, these antibodies enhance NK cell-mediated cytotoxicity and promote the activation of antigen-specific T cells and dendritic cells within the tumor microenvironment. GV20-0251 is currently undergoing Phase 1 clinical evaluation for the treatment of advanced solid tumors, where it has shown a favorable safety profile and preliminary evidence of monotherapy anti-tumor activity.
