Overview

Anti-IL-1β monoclonal antibodies are a class of biologic therapies designed to selectively bind and neutralize interleukin-1 beta (IL-1β), a potent pro-inflammatory cytokine. IL-1β plays a central role in the innate immune response and chronic inflammatory states, contributing to the pathogenesis of various autoinflammatory diseases such as cryopyrin-associated periodic syndromes (CAPS), systemic juvenile idiopathic arthritis, and gouty arthritis. By blocking the interaction of IL-1β with its receptors, these antibodies inhibit the downstream inflammatory cascade, including the production of other mediators like IL-6. The most prominent member of this class is canakinumab (Ilaris), developed by Novartis, which has also been investigated for cardiovascular risk reduction (CANTOS trial) and sickle cell disease. Other candidates in this category include gevokizumab. In preclinical research, surrogate antibodies such as the murine-specific mAb 01BSUR are frequently used to model these therapeutic effects in mice.
