Anti-IL-13Rα1 neutralizing antibodies are a class of monoclonal antibodies designed to bind and block the interleukin-13 receptor alpha 1 (IL-13Rα1) subunit. This subunit is a critical component of the heterodimeric receptor complex (formed with IL-4Rα) that mediates signaling for both IL-4 and IL-13, cytokines central to Type 2 (Th2) inflammation. By neutralizing IL-13Rα1, these biologics inhibit the activation of the JAK-STAT6 signaling pathway, thereby reducing inflammatory responses associated with allergic and fibrotic diseases. Eblasakimab is the most clinically advanced specific drug candidate within this class, currently being investigated for atopic dermatitis and other inflammatory conditions. Research also explores their role in asthma, COPD, and certain cancers where IL-13 signaling contributes to disease progression.
