Anti-IL-16 antibody is a monoclonal antibody that neutralizes interleukin-16 (IL-16), a pleiotropic cytokine functioning as a chemoattractant for CD4+ T cells, modulator of T-cell activation, and contributor to TH1-type inflammatory responses. It binds to the PDZ domain of secreted IL-16, inducing a conformational change that opens the peptide-binding groove, thereby inhibiting IL-16's pro-inflammatory activities without directly blocking the proposed CD4 binding site. Developed from research collaborations including Boston University and MRC Technology, with the 14.1 clone (humanized as c14.1) showing efficacy in preclinical models of autoimmune diseases, ischemia-reperfusion injury (IRI), sepsis-induced cardiac injury, rheumatoid arthritis, and type 1 diabetes by reducing inflammation, oxidative stress via Nrf2 pathway activation, and disease pathology.[1][2][10][12]
