Drug intelligence / Profile preview
An anti-IL-21R antagonist antibody is a monoclonal antibody designed to target and block the interleukin-21 receptor (IL-21R). By binding to IL-21R, the antibody prevents its interaction with the cytokine interleukin-21 (IL-21), thereby inhibiting downstream JAK/STAT signaling pathways (specifically JAK1, JAK3, STAT1, and STAT3). This blockade suppresses the activation, proliferation, and differentiation of various immune cells, including CD4+ T cells, B cells, and natural killer (NK) cells. Preclinical research has demonstrated that therapeutic inhibition of the IL-21/IL-21R axis can attenuate hepatic inflammation and reverse fibrosis in models of metabolic dysfunction-associated steatohepatitis (MASH), highlighting its potential as a therapeutic target for chronic inflammatory and fibrotic liver diseases.
