Anti-IL-22 blocking antibodies are a class of monoclonal antibodies designed to neutralize the cytokine interleukin-22 (IL-22). IL-22 is a member of the IL-10 family, primarily produced by Th17 and Th22 cells, and plays a critical role in mediating crosstalk between the immune system and epithelial cells, particularly in the skin, gut, and lungs. While IL-22 is essential for barrier defense and tissue repair, its dysregulation is implicated in the pathogenesis of chronic inflammatory diseases such as psoriasis and atopic dermatitis, where it promotes keratinocyte proliferation and inhibits differentiation. By binding to IL-22 and preventing its interaction with the IL-22 receptor complex (comprising IL-22R1 and IL-10R2), these antibodies aim to reduce epithelial hyperplasia and inflammatory signaling. Clinical candidates in this space have included fezakinumab (ILV-094) and BMS-981164.
