Anti-IL-34 antibodies are monoclonal antibodies designed to neutralize interleukin-34 (IL-34), a proinflammatory cytokine that serves as a ligand for the colony-stimulating factor-1 receptor (CSF-1R). Although IL-34 and CSF-1 share the same receptor, they exhibit distinct, non-redundant expression patterns; IL-34 is primarily expressed in the skin and the central nervous system (CNS). By selectively targeting IL-34, these therapies aim to modulate pathogenic myeloid cells, such as inflammatory microglia and macrophages, while potentially sparing the homeostatic functions of myeloid cells maintained by CSF-1. This approach is being investigated for the treatment of autoimmune conditions like rheumatoid arthritis and multiple sclerosis, as well as in oncology to alter the immunosuppressive tumor microenvironment by targeting tumor-associated macrophages (TAMs).
