Anti-IL-35 neutralizing antibodies are a class of therapeutic agents designed to inhibit the activity of Interleukin-35 (IL-35), a potent immunosuppressive cytokine belonging to the IL-12 family. IL-35 is a heterodimeric protein composed of the IL-12p35 and EBI3 subunits, primarily secreted by regulatory T cells (Tregs) and regulatory B cells (Bregs). It plays a critical role in maintaining immune tolerance and promoting an immunosuppressive tumor microenvironment by inhibiting the proliferation and effector functions of CD4+ and CD8+ T cells and natural killer (NK) cells. By binding to and neutralizing IL-35, these antibodies aim to restore anti-tumor immunity and enhance the efficacy of other immunotherapies, such as PD-1/PD-L1 checkpoint inhibitors. Clinical-stage assets in this space, such as WBP2101, are being evaluated for their potential to treat various solid tumors where IL-35-mediated immunosuppression contributes to treatment resistance.
