An anti-IL-4 neutralizing monoclonal antibody is a type of biologic therapy designed to bind to and neutralize interleukin-4 (IL-4), a key cytokine involved in type 2 (Th2) immune responses. By blocking IL-4, these antibodies inhibit downstream signaling pathways, such as JAK-STAT6, thereby reducing Th2 cell differentiation, immunoglobulin E (IgE) production, and alternative (M2) macrophage activation. This class of therapeutic agents has been investigated for the treatment of allergic and inflammatory conditions, such as asthma and atopic dermatitis, as well as in oncology to reprogram the immunosuppressive tumor microenvironment.
