Drug intelligence / Profile preview
An anti-IL-4 receptor alpha (IL-4Rα) monoclonal antibody is a class of biologic therapies designed to target the alpha subunit of the interleukin-4 receptor. IL-4Rα is a shared component of both the type 1 (IL-4Rα/γc) and type 2 (IL-4Rα/IL-13Rα1) receptor complexes. By binding to IL-4Rα, these monoclonal antibodies inhibit the signaling pathways of both interleukin-4 (IL-4) and interleukin-13 (IL-13), which are key drivers of type 2 (Th2-mediated) allergic inflammation. This class includes approved therapies such as dupilumab (Dupixent) and stapokibart, as well as several investigational candidates like rademikibart and MG-K10. These agents are primarily developed for the treatment of moderate-to-severe type 2 inflammatory diseases, including atopic dermatitis, asthma, chronic rhinosinusitis with nasal polyps, and prurigo nodularis.
