Anti-IL-1β antibodies are a class of monoclonal antibodies designed to neutralize interleukin-1 beta (IL-1β), a potent pro-inflammatory cytokine. IL-1β is a key mediator of the innate immune response and is involved in the pathogenesis of various autoinflammatory conditions, cardiovascular diseases, and metabolic disorders. These antibodies bind specifically to soluble IL-1β, preventing it from interacting with the IL-1 receptor type I (IL-1RI) and thereby blocking the downstream inflammatory signaling cascade. The most well-known clinical representative of this class is canakinumab, which is approved for the treatment of periodic fever syndromes, systemic juvenile idiopathic arthritis, and gouty arthritis. Additionally, anti-IL-1β therapy has been investigated for its potential to reduce major adverse cardiovascular events (as seen in the CANTOS trial) and is being studied in conditions such as heart failure with preserved ejection fraction (HFpEF) and pulmonary hypertension.
