Drug intelligence / Profile preview
An anti-IL10R monoclonal antibody is a monoclonal antibody designed to target and block the interleukin-10 receptor (IL-10R), specifically the alpha subunit (IL-10RA / CD210). IL-10 is a key immunosuppressive cytokine that plays a major role in resolving inflammation but can also promote tumor immune evasion by inducing an immunosuppressive tumor microenvironment, such as by polarizing tumor-associated macrophages (TAMs) to the pro-tumor M2 phenotype. By blocking IL-10R, these antibodies prevent IL-10-mediated signaling (which acts through the JAK1/TYK2 and STAT3 pathway), thereby restoring pro-inflammatory immune responses, enhancing T-cell and NK-cell activation, and reprogramming TAMs to an anti-tumor M1 phenotype. While widely used as preclinical research tools (such as the clone 1B1.3a) to study inflammatory bowel disease, chronic infections, and cancer immunology, anti-IL10R monoclonal antibodies are also being explored as potential therapeutic candidates in oncology and infectious diseases.
