Drug intelligence / Profile preview
An anti-IL11 monoclonal antibody is a therapeutic biologic designed to target and neutralize interleukin-11 (IL-11), a cytokine belonging to the IL-6 family. IL-11 is a key downstream mediator of TGF-beta signaling and is widely implicated in chronic inflammation, cellular senescence, and fibrogenesis across multiple organs, including the lungs, kidneys, liver, and skin. By binding to IL-11, these antibodies prevent its interaction with the IL-11 receptor subunit alpha (IL-11RA) and the gp130 co-receptor, thereby inhibiting downstream JAK/STAT (specifically JAK2/STAT3) and MEK/ERK signaling pathways. This blockade halts fibroblast activation, myofibroblast transdifferentiation, and extracellular matrix deposition. Anti-IL11 monoclonal antibodies are being investigated for the treatment of various fibro-inflammatory conditions, such as idiopathic pulmonary fibrosis (IPF), chronic kidney disease, pathological scarring (including keloids and hypertrophic scars), and age-related multimorbidities.
