Anti-IL13Ra2 refers to a class of therapeutic agents, including monoclonal antibodies, antibody-drug conjugates (ADCs), and CAR-T cell therapies, designed to target the Interleukin-13 receptor subunit alpha-2 (IL13Rα2). IL13Rα2 is a high-affinity monomeric receptor for IL-13 that is significantly overexpressed in various cancers, most notably glioblastoma multiforme (GBM), diffuse intrinsic pontine glioma (DIPG), and melanoma, while showing minimal expression in normal tissues. Because IL13Rα2 lacks a cytoplasmic signaling domain and acts as a decoy receptor, it is primarily utilized as a tumor-specific antigen for targeted delivery of cytotoxic payloads or for directing immune effector cells. Several specific candidates have entered clinical development, such as the ADC ABBV-066 and various CAR-T constructs developed by institutions like City of Hope.
