Anti-IL1RAP antibodies are a class of biologic therapeutics designed to target the Interleukin-1 Receptor Accessory Protein (IL1RAP, also known as IL1R3). IL1RAP is a critical co-receptor for multiple pro-inflammatory cytokine subfamilies, including IL-1, IL-33, and IL-36, making it a central node in inflammatory signaling. By blocking IL1RAP, these antibodies can simultaneously inhibit multiple signaling pathways involved in cancer progression and autoimmune diseases. In oncology, IL1RAP is often overexpressed on cancer cells and leukemic stem cells (LSCs), and its inhibition can trigger antibody-dependent cellular cytotoxicity (ADCC) or block growth-promoting signals. Several companies are developing candidates in this space, including Cantargia (nadunolimab, CAN10), Singlomics (DXP-006, DXP-106), and Cellerant Therapeutics.
