Anti-integrin alpha5 monoclonal antibodies are a class of therapeutic agents designed to target and inhibit the integrin alpha-5 (ITGA5) subunit. ITGA5 typically dimerizes with the beta-1 subunit to form the α5β1 integrin, also known as the fibronectin receptor. This integrin complex is a critical mediator of cell-extracellular matrix interactions, playing a pivotal role in cell adhesion, migration, proliferation, and survival. In oncology, α5β1 is frequently overexpressed on both tumor cells and the vascular endothelium of the tumor microenvironment, where it promotes tumor angiogenesis and metastasis. By binding to the ITGA5 subunit, these monoclonal antibodies block the interaction between α5β1 and its primary ligand, fibronectin, thereby inhibiting angiogenic signaling and tumor progression. Clinical development in this space has included assets such as volociximab (M200) and PF-04605412, which were investigated for various solid tumors, including ovarian cancer, renal cell carcinoma, and melanoma.
