Anti-integrin beta1 antibodies are a class of monoclonal antibodies that target and inhibit integrin beta-1 (also known as ITGB1 or CD29), a cell surface receptor that serves as a common beta subunit for various alpha integrins (e.g., alpha-4, alpha-5, alpha-L). By blocking the interaction between integrin beta-1 and its extracellular matrix ligands (such as fibronectin and laminin) or cell-surface ligands (such as VCAM-1 and ICAM-1), these antibodies disrupt critical cell-cell and cell-matrix adhesion signaling. In oncology, integrin beta-1 blockade has been shown to reduce the survival of T-cell acute lymphoblastic leukemia (T-ALL) cells by interrupting support from tumor-associated myeloid cells and inhibiting IGF1R activation. In glioblastoma, targeting integrin beta-1 interferes with the trafficking and pro-tumorigenic functions of immunosuppressive monocytic myeloid-derived suppressor cells (mMDSCs). While frequently used as research reagents, therapeutic candidates such as OS2966 have entered clinical development for high-grade gliomas and other solid tumors.
