Anti-interleukin-33 (anti-IL-33) refers to a class of monoclonal antibodies designed to neutralize interleukin-33, an alarmin cytokine belonging to the IL-1 family. IL-33 is constitutively expressed in the nuclei of epithelial and endothelial cells and is released upon cell injury or stress, acting as a potent initiator of type 2 innate and adaptive immune responses. By binding to soluble IL-33, these antibodies prevent its interaction with the ST2 (IL-1RL1) receptor complex on cells such as mast cells, eosinophils, and Type 2 innate lymphoid cells (ILC2s). This blockade inhibits the production of Th2 cytokines (IL-4, IL-5, and IL-13), thereby reducing airway hyperresponsiveness, mucus hypersecretion, and tissue eosinophilia. Clinical development of anti-IL-33 agents focuses on inflammatory conditions including asthma, chronic obstructive pulmonary disease (COPD), and atopic dermatitis.
