Drug intelligence / Profile preview
An anti-JAG1 antibody is a monoclonal antibody designed to target and inhibit Jagged-1 (JAG1), a key ligand in the Notch signaling pathway. JAG1 is overexpressed in various cancers and in the bone marrow microenvironment (specifically on osteoblasts) of patients with myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML), where it drives oncogenic Notch1 signaling in hematopoietic stem cells. A prominent candidate in this class is ATI-D1 (also known as D1), a humanized antagonist monoclonal antibody developed by Alanis Therapeutics (a spinout of AvantGen) in collaboration with Columbia University. ATI-D1 blocks the interaction between osteoblastic JAG1 and Notch1 on leukemic stem cells, reversing cytopenias, relieving myeloid differentiation blocks, and eliminating blasts in preclinical models of MDS and AML. Other anti-JAG1 antibodies, such as Genentech's aJ1.b70, have been investigated in solid tumors like hepatocellular carcinoma.
