Anti-JL1 monoclonal antibody-saporin conjugate is an experimental immunotoxin (antibody-drug conjugate) being investigated for the treatment of acute leukemia, including acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL). The drug consists of the JL1 monoclonal antibody, which targets a specific developmentally regulated epitope of CD43 (sialophorin) expressed on leukemia blasts across T, B, and myeloid lineages, but absent on normal peripheral blood cells and hematopoietic stem cells. This antibody is conjugated to saporin, a type I ribosome-inactivating protein (RIP) derived from Saponaria officinalis. Upon binding to the target cell and subsequent internalization, saporin catalyzes the depurination of the sarcin/ricin loop of the 28S ribosomal RNA, leading to the irreversible inhibition of protein synthesis and induction of apoptosis. The conjugate has demonstrated potent, dose-dependent anti-leukemic activity in vitro and in preclinical models.
